雌激素对脑缺血时bcl-2蛋白表达的影响 被引量:1

摘要 目的 研究雌激素对缺血性脑损害的神经保护作用是否与bcl- 2蛋白的表达有关。方法 采用大鼠大脑中动脉闭塞制成局灶性脑缺血模型。在缺血 2h ,再灌注 2 2h后立即断头取脑 ,应用TTC染色和免疫组化方法 ,观察脑梗死体积及bcl- 2蛋白的表达。结果 雌激素用药组较对照组脑梗死体积显著缩小 (P <0 0 5 ) ;bcl- 2蛋白的表达较对照组明显增强(P <0 0 5 )。结论 雌激素上调大鼠脑缺血时bcl- 2的表达很可能是其具有神经保护作用的机制之一。 Objective To study the protective effect of estrogen against neuronal injury and the possible roles of alteration in the expression of bcl-2 following cerebral ischemia.Methods Female Wistar rats were all ovariectomized.Brain ischemia was induced by left middle cerebral artery occlusion(MCAO)for 2 hours,at 22 hours of reperfusion,all animals were killed.Infarction volume was determined by 2,3,5-triphenyltetrazlium chloride(TTC)staining and image analysis.The expression of bcl-2 was analyzed by immunohistochemical method.Results Infarction volumes were reduced significantly in all estrogen-treated groups (P<0.05);Estrogen increased the expression of bcl-2(P<0.05).Conclusions Estrogen effectively attenuated ischemic brain injury and increased the expression of neuronal bcl-2 in the ischemic brain,indicating that the neuroprotective of estrogen was associated with up-regulation of bcl-2 in cerebral ischemia.
