川芎嗪对兔脊髓缺血损伤依剂量的治疗作用(英文) 被引量:5

摘要 目的 研究川芎嗪 (TMP)注射液对兔脊髓缺血损伤治疗作用的剂量效应关系 .方法  2 6只新西兰雄性大白兔 ,随机分成四组 :对照组即 C组 (n=6 ) ,单组缺血再灌注 ;T1 组(n=6 )、T2 组 (n=8)和 T3组 (n=6 ) ,分别在肾下主动脉阻断2 0 min后开放时恒速静脉泵入 TMP注射液 15 ,30和 6 0 mg· kg- 1 .采用肾下主动脉 (IRA)阻断法造成脊髓缺血 (2 0min) /再灌注模型 ,观察再灌注后 4,8,12 ,2 4和 48h神经功能评分 (neurologic deficit score) ,并于 48h处死动物取脊髓(L5 - L7)制标本行病理组织学观察 .结果 用不同剂量TMP治疗的 T1 ,T2 和 T3组在各时间点神经功能评分和再灌注 48h脊髓前角正常神经元数均明显高于 C组 (P<0 .0 1) ,且 T2 组和 T3又明显高于 T1 组 (P<0 .0 5 ) ,而 T2 组和 T3组无显著差异 ,神经功能评分与其对应脊髓前角正常神经元计数之间有显著相关性 (r=0 .776 ,P<0 .0 1) .结论  TMP注射液对脊髓缺血损伤有治疗作用 。 AIM To investigate the dose response relation between tetramethylpyrazine (TMP), an intravenous drug made from traditional Chinese herbs, and the neurologic injury related to spinal cord ischemia and reperfusion in rabbits. METHODS Twenty six male New Zealand white rabbits were anesthetized with isoflurane and spinal cord ischemia was induced for 20 min by infrarenal aortic occlusion. Animals were randomly allocated to four groups. Group C received no pharmacologic intervention. Group T 1, Group T 2 and Group T 3 received 15, 30 and 60 mg·kg -1 TMP intravenous infusion respectively within 30 min at the end of ischemia. Neurologic status was scored by the Tarlov criteria (in which 4 is normal and 0 is paraplegia) at 4, 8, 12, 24, and 48 h after reperfusion. All the animals were sacrificed at 48 h after reperfusion and the spinal cords (L 5-L 7) were removed immediately for histopathologic study. RESULTS All the animals survived the experiment. The neurologic scores at each timepoint and the number of normal neurons in the anterior spinal cord 48 h after reperfusion of Groups T 1, T 2, and T 3 were significantly more than those of Group C ( P <0.01). Those of Group T 2 and T 3 were markedly more than those of GroupT 1, but no difference was found between Groups T 2 and T 3. There was a strong correlation between the neurologic scores and the number of normal neurons in the anterior spinal cord. CONCLUSION TMP may signifi cantly reduce neurologic injury induced by spinal cord is chemia and reperfusion in a manner with a certain range of dosage.
