摘要
探讨细胞间粘附分子 1(ICAM 1)及中性粒细胞 (Neu)炎性因子巨噬细胞炎症蛋白 2 (MIP 2 )、髓过氧化物酶(MPO)在COPD大鼠模型气道炎症中的作用。制作COPD大鼠模型 (模型组 ) ,布地奈德组及异丙托品组分别于制作模型后雾化吸入此二药物 ,对照组为空白对照。用免疫组化及原位杂交法检测ICAM 1在肺内的表达 ,用ELISA法检测血清、支气管肺泡灌洗液 (BALF)及肺组织MIP 2及肺组织MPO。模型组及异丙托品组支气管粘膜上皮和肺毛细血管内皮细胞ICAM 1的表达强度及BALF中Neu数显著高于对照组 ,模型组BALF和肺组织MIP 2、肺组织MPO显著高于对照组。布地奈德组上述指标均显著低于模型组。气道内中性粒细胞的聚集与ICAM 1表达上调及MIP 2特异性趋化作用有关 ,MPO升高表明Neu的显著活化 ;吸入皮质激素可能通过下调ICAM 1的表达 ,减轻气道炎症。
To observe the ICAM 1、Macrophage inflammatory protein 2(MIP 2) and Myoloperoxidase(MPO) in bronchopulmonary tissues of a COPD rat model , as well as the effects of Budesonide or Atrovent on them, the expression of ICAM 1 on bronchopulmonary tissues was detected by means of immunohistochemistry and in situ hybridization;MIP 2 and MPO were measured by ELASA The expression of ICAM 1 on pulmonary capillary endothelial and airway epithelial cells was significantly enhanced in the model group and Atrovent group compared with control group Significant increases in total leukocyte numbers and neutrophils in BALF were found in model group than those in control group Significant decrease in ICAM 1 expression on endothelium and epithelium and in BALF leukocyte numbers was found in Budesonide group compared to those of model group The level of MIP 2 in BALF and lung tissue and MPO in lung tissue homogenate in model group were significantly increased than those of control group ICAM 1、MIP 2 and MPO may be involved in the process of attachment, chemotaxis and activation of neutrophils in airway inflammation in COPD Inhaled corticosteroide might have some effects on limiting the airway inflammation
