摘要
采用碘化丙啶和FITC TUNEL标记法 ,经流式细胞仪和透射电镜观察As2 O3 对肺癌A5 49细胞株的凋亡诱导作用和形态学变化。结果发现 1及 2 5 μmol/LAs2 O3 对肺癌A5 49细胞株 2 4h的凋亡诱导率为 2 2 80 %及 30 15 % ,48h的凋亡诱导率为 41 75 %及 6 0 0 4% ,72h的凋亡诱导率为 5 6 38%及 73 30 % ,均分别高于相应时间点的 5 FU的作用 ;透射电镜下见细胞体积缩小 ,核膜完整 ,核染色质聚集于核膜边缘的典型凋亡细胞。结果表明 ,As2 O3 能够诱导肺癌A5
To study whether arsenic trioxide (As 2O 3) will induce apoptosis of lung cancer cells, apoptosis rate and morphologic changes of A549 cells induced by As 2O 3 were observed under flow cytometry(FCM) assays and transmission electric microscopy(TEM) It was found that apoptosis rate induced by As 2O 3 of 1、2 5μmol/L was 22 80%,30 15% at 24 hours; 41 75%,60 04% at 48 hours and 56 38%,73 70% at 72 hours, respectively, and they were all higher than those acted by 5 Fu Typical apoptotic cells were seen under TEM These results indicated that As 2O 3 induced apoptosis in lung cancer cells line A549, which provides a new path to the treatment of lung cancer
