摘要
从沿海滩涂土壤、海鱼消化道、海水和海底污泥等自然环境中采集样品 34个 ,从中分离获得能降解甲壳素的微生物 1 1 4株 ,经过初筛和复筛 ,从中获得产酶能力较高的菌株 C43,用比色法测定其酶活 ,发酵液粗酶活达 0 .1 1 5u。初步鉴定 C43菌株为噬纤维菌科、噬纤维菌属。暂命名为噬纤维菌 C43( Cytophaga.sp C43)。研究表明 :噬纤维菌 C43产生的甲壳素水解酶为诱导型。其产酶的最佳条件为 :起始 p H6.0 ,温度 2 8℃ ,摇瓶转速 2 0 0 r/min,发酵周期为 60 h。
chitinase bacterium strains were isolated from 34 samples collected from sea water, sea soils, and alimentary tract of sea\|fish, etc. Strain C43, identified as Cytophaga.sp , held the chitinase production rate of 0.115 u with the highest activity in all these strains. Study showed that the chitinase produced by Cytophaga.sp C43 was induced. The optimal conditions for chitinase production were the initial pH value of 6.0, the culture temperature of 28℃, and the stirring rate of 200 r/min for 60 h.
