摘要
以 3份人参果栽培品种为供试材料 ,比较了启动培养基中激素浓度和不同的剥茎尖时期对茎尖成活率的影响、不同脱毒方法对脱毒率的影响、细胞分裂素对脱毒苗繁殖系数的影响以及生长素对脱毒苗茎杆粗细的影响。结果表明适宜人参果茎尖培养的启动培养基为含 1.0mg/L 6-BA和 0 .0 5mg/LNAA的MS培养基 ;人参果脱毒剥取茎尖的最佳时期来自组培苗经 35℃高温热处理 1月后剥取的茎尖 ;采用连续多代 35℃高温热处理加剥茎尖培养的方法 ,获得了人参果脱毒试管苗 ;快繁培养基中 1.5mg/L与 0 .5mg/L两种浓度的 6 -BA交替使用既提高了繁殖系数又有效地控制了玻璃苗现象的发生 ;培养基中添加 0 .0 5mg/L的NAA ,可使脱毒苗生长健壮。
The effects of hormone combinations in media on survival apical meristems rate and growth of virus-free plant, different isolation of apical meristems on their survival rate and on percentage of virus-free plant were studied by using 3 cultivars. The results showed that MS medium supplemented with 1.0mg/L 6-BA and 0.05mg/L NAA was suitable for stem tips growth, the way of alternation of 1.5mg/L and 0.5mg/L 6-BA combined with 0.05mg/L NAA was able to increase propagation rate of virus-free plant, the optimal effect of taking off virus was isolating apical meristems after treating plant with 35℃ for 1 month.
