GUS(β—glucuronidae)基因在水稻植株中的表达

摘要 采用花粉管通道法,将带GUS基因的PBI121质粒DNA导入栽培稻农垦58。用荧光法检测GUS基因在水稻植株中的表达以及根据Dot blotting和Southern blotting的结果分析,揭示了经花粉管通道途径导入的外源DNA整合到了转基因植株的基因组中,并在某些转基因植株中得到表达。本文还对转基因植株后代出现的各种变异型进行了探讨。 Sucess in the introduction of bacterial - glu-curonidase ( GUS gene into the cultevar rice LONGKEN58 by pollentube pathway and glu-curonidase activity was detected in extracts of the 3F1 transtormed rice leaves. PBI121 plasmids DNA were labed with Biotin - psoralen. the result of Dot hybridization:the 7 DNA samples among 100 DNA samples were positive reaction. Perparing a - p32 labed PBI121 plasmids DNA by Nick - Translation method. The transformed rice genomic DNA is digested with restriction enzyme BamHI.and the resulting fragmemts were separted according to size by elec-tronphoresew through an 1. 4% agrose gel, the DNAwas then denatured in situ and transferred from the gel to a nitrocellulose filter.The DNA probe,and appear 2 bands in the x - film by autoradiography. GUS gene have integrated in the genome of the transformed rice plants by pollen tube pathway. The characters in trans - formed rice plants appeared variation most of them can produce mormally seeds.
