用RAPD技术鉴定烤烟抗赤星病基因连锁标记 被引量:12

摘要 用49个RAPD引物对烤烟13个抗赤星病品种和15个感赤星病品种基因组DNA进行RAPD分析,找到了一个和烤烟抗赤星病表型密切相关的DNA片段,此片段长760bp,命名为S220-760。通过对长勃黄和净叶黄的RAPD分析表明,S220-760为13个抗病品种所共有和15个感病品种没有。因此,S220-760可作为烤烟抗赤星病基因紧密连锁的RAPD标记。这为下一步钓取烤烟赤星病基因的工作打下了基础。 The genome DNA isolated from 13 brown spot disease-resistant and 15 susceptible germplasm of flue-cured tobacco were amplified with 49 random primers by RAPD technique. A DNA fragment named S220-760, which is closely related to brown spot disease-resistance phenotype, was detected in 13 resistant cultivar. RAPD analysis of changbo-huang and jingyehuang indicates that S220-760 is only existent in jinyehuang of brown spot disease-resistant. By this token, S220-760 may be a part or flank sequence of brown spot disease-resistant gene, which may result in a possibility for separation of brown spot disease resistant gene from flue-cured tobacco.
