摘要
目的:建立RP-HPLC测定白花前胡根中白花前胡丙素(Pd-Ia)和白花前胡丁素(Pd-Ⅱ)的含量。方法:采用Shim-Pack CLC-ODS柱,甲醇-水(79:21)为流动相,流速为 1.0ml·min-1,以 Pd-Ⅰa、Pd-Ⅱ为对照品,外标法峰面积定量,紫外检测(λ=323nm)。结果:Pd-Ⅰa、Pd-Ⅱ线性范围分别为0.08~0.8μg和0.064~0.28μg,r=0.9999和r=0.9999,平均回收率分别为 98.90%和 98.91%。结论:方法准确,简便,灵敏度高,快速,重复性好。
OBJECTIVE: To establish HPLC method for determining the contents of Pd-Ⅰa and Pd-Ⅱ in root of Peucedanum Praeruptorum. METHODS: The powdered samples was extracted with CHCl3 and CH3OH, the filtrate was analyzed by RP-HPLC, using ODS column, UV detection at 323 non, CH3OH-H2O(79:21 ) as mobile phase and Pd-Ⅰa, Pd-Ⅱ reference standard as external standard. RESULTS: The calibration curves showed good linearity over the range of 0.08 - 0.8 μg and 0.064 - 0.28 μg, r = 0.9999 and 0. 9999, the average mrecoveries were 98. 90% and 98. 91%, respectively. CONCLUSION: The method was simple, sensitive and reproducible, and can be used for the quality control of Chinese medicine Qianhu.
