摘要
目的 观察电针对应激大鼠胃粘膜血流量与血浆内皮素 (ET)、一氧化氮 (NO)、降钙素基因相关肽 (CGRP)的影响及其相互关系 .方法 将 32只 SD大鼠平均分为空白对照组、应激组、电针后应激组和应激后电针组 ,每组 8只 .利用激光多普勒血流仪测定胃粘膜血流量 ,用放免分析法和生化法测定各组大鼠血浆 ET,CGRP和 NO水平的变化并计算各组溃疡指数 (UI) .结果 应激组较对照组比较胃粘膜血流量相对数值下降 (6 9.0± 10 .6 ) m V→ (5 1.5± 5 .1) m V,P<0 .0 1. UI增加 (1.1± 0 .4)→ (2 8.0± 4.1) ,P<0 .0 1.血浆 NO水平下降 ,(2 2 .7± 3.8) μmol· L- 1 → (18.8± 5 .2 ) μmol·L- 1 ,P<0 .0 5 . ET水平上升 (140± 17) ng· L- 1 → (177± 2 3)ng· L- 1 ,P<0 .0 5 .在电针后应激组和应激后电针组 ,与应激组比较 ,胃粘膜血流量上升 (5 1.5± 5 .1) m V→ (6 6 .5± 7.4) m V,(6 4.9± 5 .8) m V,P<0 .0 1,UI减少 ;(2 8.0± 4.1)→ (19.0± 2 .3) ,(19.4± 2 .8) ,P<0 .0 1.血浆 NO水平上升 ,(18.8± 5 .2 )μmol· L- 1 → (2 3.3± 4.1)μmol· L- 1 ,(2 2 .9± 4.1) μmol· L- 1 ,P<0 .0 5 ;CGRP水平上升 ,(145± 6 ) ng· L- 1→ (184± 2 2 ) ng· L- 1 ,P<0 .0 5 ;ET水平下降 ,(177±2 3)
AIM To explore the effects of electroacupuncture (EA) on gastric mucosal blood flow (GMBF) and plasma levels of endothelin (ET), nitric oxide (NO), and calcitonin gene related peptide (CGRP) in stress rats. METHODS 32 SD rats were randomly divided into 4 groups: control group, stress group, EA plus stress and stress plus EA groups. GMBF was measured directly by laser Doppler flowmeter (LDF). The plasma levels of NO and CGRP were measured by radioimmunoassay methods and the level of NO by biochemical methods. The extent of gastric mucosal damage was evaluated by ulcer index (UI), according to the Guth exponential method. RESULTS In the stress group, the mean value of GMBF decreased (69.0±10.6) mV→(51.5±5.1) mV, P <0.05; UI increased (1.1±0.4)→(28.0±4.1), P <0.01, the level of NO decreased (22.7±3.8) μmol·L -1 →(18.8±5.2) μmol·L -1 P <0.05, but ET increased (140±17) ng·L -1 →(177±23) ng·L -1 , P <0.05; as compared with those of control group. In the EA plus stress and stress plus EA groups compared with stress group,GMBF increased (51.5±5.1) mV→(66.5±7.4) mV, (64.9±5.8) mV, P <0.01. UI decreased (28.0±4.1)→(19.0±2.3), (19.4± 2.8 ), P <0.01. The levels of NO increased (18.8±5.2) μmol· L -1 →(23.3±4.1) μmol·L -1 , (22.9±4.1) μmol·L -1 , P <0.05; CGRP increased (145±6) ng·L -1 →(184±22) ng·L -1 , P <0.05; but ET decreased (177±23) ng·L -1 →(134±8) ng·L -1 , (140±20) ng·L -1 , P <0.05. Except for CGRP in stress plus EA group. CONCLUSION EA at zusanli point evidently increases GMBF, and protects gastric mucosa. This protection may be connected with the changes of plasma levels of ET, NO and CGRP.
