摘要
为寻找简便可行的脑囊虫病诊断方法,将2株抗猪囊尾蚴囊液抗原McAb,一个滴于硝酸纤维素膜上,另一与胶体金结合,建立了检测脑囊虫病患者脑脊液(CSF)中循环抗原(CA)的金标免疫渗滤法(DIGPA)。本法中抗原、抗体通过渗滤在膜上进行反应,10分钟内即可用肉眼观察结果.以该法检测48例脑囊虫病患者CSF、26份健康人CSF及23例非囊虫病患者CSF。对检测样本作了沸浴处理,处理前后的48例脑囊虫病患者CSF阳性检出率分别是77.08%和93.75%,处理前后的对照CSF阴性符合率均未改变.26例健康人CSF,阴性符合率为96.15%;23例非囊虫病患者CSF,1例出现阳性反应,交叉反应率为4.35%。该方法可检出最小抗原量为0·5μg/L沸治处理可提高脑囊虫病人CSF的CA阳性检出率. DIGFA敏感、特异、重复性好,操作简便,无需特殊设备,可用于囊虫病的诊断。
In order to establish a rapid, sensitive and specific method for diagnosie of neurocysticercosis, two McAbs against cystic fluid antigen of Cysticercus cellulosae, one absorbed on nitrocellulose membrane and another conjugated to colloidalgold, wer used to develop a dot immunoglod filtation assay (DIGFA) for the detection of circulating antigen (CA) in cerebrospinal fluid (CSF) of pa- tients with neurocysticercosis. Antigen-antibody reaction was completed on the meinbrane during the filtration process. The result could be read with the naked eye within ten minutes. CSF of 48 cases with neurocysticercosis, 26 samples from normal people and 23cases with other diseases of the central nervous wystem as detected. All samples Were treated by boiling water bath. Before and after the treatment on CSF, the posistive rates of CA in CSF of 48 cases with neurocysticercosis were 77. 08% and 93. 75% respectively while the negative coincidence rates of control CSF remained no change. Out of 26 samples of CSF from normal population only one showed positive. 23 samples of CSF from patients with other diseases of the central nervous were negative except one. The results showed that the positive rate of CA in CSF of patients with neurocysticercosis could be increased by treating on CSF with boiling water bath. The minimal detectable level of DIGFA was 0. 5 μg/mL. It suggested that this assay was sensitive, specific and simple and suitable for diagnosis of neurocysticercosis.
