摘要
通过形态观察和 16SrDNA序列同源性比较 ,将一株产耐高温碱性蛋白酶菌株GXD990 1初步鉴定为枯草芽胞杆菌 (Bacillussubtilis)。对GXD990 1菌株产蛋白酶的培养条件进行分析表明 ,最佳产酶培养基为 :麦芽糖 3%、NH4 H2 PO4 0 3%、MnCl2 0 0 5%、KCl0 0 5%、Fe2 (SO4 ) 30 0 0 1%、K2 HPO4 0 1% ,pH11 2。在 37℃培养 6 0h后 ,蛋白酶活力达到546
On the basis of the observing of modality and the 16S rDNA sequence analysis ,GXD9901,a thermostable alkaline protease producing strain,was identified as Bacillus subtilis. The optimum condition for the protease production consisted of 3% maltose, NH 4H 2PO 4 0.3%,MnCl 2 0.05%,KCl 0.05%,Fe 2(SO 4) 3 0.001%, K 2HPO 4 0.1%, pH?11.2. After fermentation at 37℃ for 60h,the enzyme activity was about 5?465?U/mL of fermentation liquor。
