谷子及其近缘野生种酯酶同工酶分析(Ⅱ) 被引量:3

摘要 采用聚丙烯酰胺凝胶电泳技术 ,对谷子及其 8个狗尾草野生种进行酯酶同工酶分析。结果表明 :谷子及其不同野生种种间同工酶谱差异性主要表现为大多数酶带的差异 ,酯酶同工酶的酶带条数与各种间的倍性水平无明显相关关系。谷子、西藏 1 90和青狗尾草 3个物种间的亲缘关系最为密切 ,这 3个物种与法氏狗尾草、断穗狗尾草表现出较近的亲缘关系 ,轮生狗尾草的 2个种居中 ,而金色狗尾草、皱叶狗尾草之间以及与上述几个物种间的亲缘关系较远。轮生狗尾草二倍体种与轮生狗尾草四倍体种表现出较近的亲缘关系。将前人的细胞遗传学研究结果与本试验结果相结合进行分析 ,发现谷子染色体组具有 4条酯酶同工酶标志带 ,通过这 4条标志带可初步判定在狗尾草属某一野生种中谷子 (青狗尾草 )染色体组的存在。结果还表明 ,青狗尾草同工酶谱的类型 与法氏狗尾草、断穗狗尾草的起源有关。 Using polyacrylamide gel electrophoresis (PAGE),analytical results are reported in this paper on esterase zymograms in foxtail millet and related species. The results indicated that introspecific differentiation of esterase zymogram patterns is main due to the differentiation of most number isozyme bands. The number of EST isozyme bands is not related to ploid of corresponding species distinctly. In many wild species, S.viridis and Tibet 190 are the closest to foxtail millet. S.faberii and S.arenaria take the second place, and S.verticilata and S.verciformis the third place, S.glauca and S.plicata are distanct between them and to the above mentioned species. In addition, S.verticilata is Closer to S. verticiformis on relationship. The results of this experiment, combining the former cytogenetical investigations,showed that the genome of foxtail millet have 4 EST isozyme bands identified. Based on these bands, the genome of foxtail millet (and S.viridis ) in other species of Setaria will be determined. This experiment evidences also showed that green foxtail millet ( S. viridis ) is divided into 3 types according to esterase patterns, in which that the type III may be ancestor of S.faberii and S.arenaria in the most possibility.
