原发性高血压患者c-myc基因表达及其与血管紧张素Ⅱ和内皮素的关系 被引量:1

A study on the expression level of proto-oncogene c-myc and its relationships with plasma AngⅡ and ET in patients with essential hypertension

摘要 目的 :研究原癌基因 c- m yc在原发性高血压 ( EH)患者外周淋巴细胞中的表达及与血浆中血管紧张素 ( Ang )和内皮素 ( ET)之间的关系。方法 :采用 PCR技术检测 EH患者 5 5例 ( EH组 , 期 17例 , 期 2 4例 , 期 14例 ) ,正常对照组 2 4例淋巴细胞中的原癌基因 c- m yc的表达水平 ,采用放射免疫法测定各组血浆中Ang 、ET含量。结果 :EH组 c- m yc的表达水平明显高于对照组 ( P <0 .0 1) , 、 期较 期增高 ( P <0 .0 1)。EH组血浆 Ang 、ET高于对照组 (分别 P <0 .0 5和 <0 .0 1) ,EH组 3期之间相比差异无显著性意义 ( P >0 .0 5 ) ;EH患者原癌基因 c- m yc的表达程度与血浆中 Ang 、ET含量呈正相关 ( r =0 .86,P <0 .0 1)。结论 :从分子水平探讨 EH的发病与 c- m yc基因表达增高有关。c- myc的表达水平与血浆中 Ang 。 Objective:To measure the altenation of the expression of lymphocytic proto oncogenes c myc mRNA and its relationships with plasma angiotensinⅡ(AngⅡ) and endothelin(ET)in patients with essential hypertension (EH).Method:Fifty five patients with EH were divided into 3 groups(17 cases in stageⅠ, 24 cases in stageⅡ, 14 cases in stage Ⅲ).Twenty four normal individuals matched in age and heart ratio and body mass index were served as normal controls(NC).The expression of c myc was measured by Polymerase Chain Reaction(PCR) and AngⅡ, ET were detected by radioimmunoassay.Result:①The level of mRNA of c myc in EH was significantly higher than that in NC group(P< 0.01 ), which was more obvious in stage Ⅱ, Ⅲ EH than in stageⅠ.②Plasma AngⅡ, ET in EH were higher than those in NC group(P< 0.05 and P< 0.01 ).The difference of AngⅡ and ET had no statistical significance among stage Ⅰ, Ⅱ and Ⅲ EH (P> 0.05 ). Conclusion:These data strongly indicate that higher expression of c myc mRNA in patients with EH may play an important role in hypertensive pathogenesis.
