壳多糖拮抗TNF促进兔主动脉内皮细胞与单核细胞粘附作用

摘要 目的:研究肿瘤坏死因子(TNF)对体外培养的免主动脉内皮细胞(RAEC)与单核细胞(MC)粘附的影响及壳多糖的拮抗作用。方法:通过测定MC髓过氧化物酶活性的方法,对粘附于RAEC单层上的MC进行定量。结果:TNF在5-500U·ml-1时诱导RAEC与MC的粘附,TNF浓度为250U·ml-1时促粘附达最大值(81.98%)。TNF与内皮细胞共育1h,即可明显促进粘附作用(66.8%),4h时达最大刺激效应(77.5%)。壳多糖在浓度为20-2000mg·L-1时,呈剂量依赖性地拮抗TNF诱导的粘附作用,在2000mg·L-1时抑制率达22.99%。结论:壳多糖具有抑制由TNF诱导的RAEC与MC粘附的作用。 OBJECTIVE.. To study inhibitory effects of chitosan on tumor necrosis factor(TNF)induced adhesion reaction of cultured rabbit aortic endothelial cells (RAEC) with monocytes(MC) in vitro. METHODS: The assay was based on the measurement of the myeloperoxidase achvity in monocytes that had been adhered to RAEC layers. RESULTS: At concentration from 5 to 50()U.ml-1,TNF was shown to induce adhesion reaction of cultured RAEC with MC. The effect of TNF reached maximum (81 .98%) at 250U·ml-l. After lh incubation of RAEC, addition of TNF significantly stimulated the adhesion reactin of RAEC with MC(66.8%).After 2h incubation the effect reached maximum(77.5%),then the effect slightly decreased. At concentration from 20 to 2000mg·L-1, chitosan was found to antagonize induced adhesion reaction. Its maximal rate of inhibition was 22.99% at 2000mg.L-1. CONCLUSION: Chitosan may decrease the TNF induced adhesion reaction of RAEC with MC.
