摘要
以羧甲基纤维素钠(CMC)为底物评价纤维素酶的活性,研究了不同条件下静磁场对酶的活性及构象的影响。9℃,pH4.0条件下磁化,其活性及荧光光谱均不见明显变化;但若改变反应时的pH,可使活性增加达16.4%;并发现磁处理后的酶有滞后效应及可逆性。较高温度下,磁化处理酶液,其活性及构橡均发生不同程度的变化,且对其抑制动力学表现为混合型抑制模式;其构象变化也基本上随失活的增加荧光强度增大。
Cellulase actlvity is measured on sodium carboxymethyl -cellulose substrate by Dinitrosalicylic acid(DNS) method. No obvious effect is observed when magnetic field on cellulase at 9℃,pH4.0. Once change the reaction pH, the activity will be increased by 16. 4%,the delaying and reversible effect is also observed- At 60℃, the effect on the activity and conformation changes at different extent. A Lineweaver-Burk plot of the reaction shows the typical pattern of mixed-type inhibition. It seems that the intensity of the fluorescence increases with the decrease of the activity.
