非特异性酯酶在蚊虫不同种类和不同发育期的变化 被引量:3

摘要 目的 研讨非特异性酯酶 (Non -specificesterase,NSE)在蚊虫不同种类和不同发育期的变化 ,为应用生化方法检测蚊虫抗药性及科学合理地进行蚊虫化学防治提供依据。方法 以β -乙酸萘酯为底物 ,坚固蓝B盐为显色剂 ,用微量滴定板法测定单个蚊虫NSE活力。结果 显示 4种敏感蚊虫的Ⅳ龄幼虫NSE活力有非常显著性差异 (F =2 2 9 9842 ,P <0 0 1) ,3日龄雌成蚊NSE活力亦有非常显著性差异 (F =37 1873,P <0 0 1) ;敏感品系和抗DDVP品系淡色库蚊NSE活力水平在不同发育期有变化 ,Ⅰ~Ⅲ龄幼虫较低 ,Ⅳ龄幼虫最高 ,蛹期下降 ,成虫期较高 ,各发育期抗DDVP品系NSE活力均高于敏感品系。结论 测定NSE检测蚊虫抗药性可选择Ⅳ龄幼虫或 3日龄未吸血雌成蚊 ,不同种类蚊虫应分别制定判断标准 ;蚊虫化学防治应选择低龄幼虫施药。 Objective Non specific esterase(NSE)was measured for different species of mosquitoes at their different development,to provide insight into rational use of chemical insecticides and biochemical detection of insecticide resistance.Methods Microplate test of NSE was performed using β-Naphthyl acetate as the substrate and fast blue B salt solution as the colour agent.Results NSE activity varied significantly among 4 species of susceptible mosquitoes in both their larval stage and 3-day-old females.DDVP-resistant strain had higher NSE level than susceptible strain,and in both case,adult showed higher NSE level than Ⅳ-instar larvae,followed by Ⅰ-Ⅲ instar larvae.Conclusion Detection of NSE using Ⅳ-instar larvae or 3-day-old females can be used for insecticide resistance detection but with different criteria for different species of mosquitoes.
