三氧化二砷诱导肺癌细胞凋亡的体外实验研究

Preliminary Study on Lung Cancer Cell Apoptosis lnduced by Arsenic Trioxide ln vitro

目的 观察三氧化二砷体外作用于肺癌 A549细胞株所致的凋亡作用及形态学改变 ;方法 采用 MTT法、透射电镜法及 TUNEL荧光染色法观察不同浓度 As2 O3 对肺癌 A549细胞株的作用效果 ;结果 1及 2 .5μmol/ L As2 O3 对肺癌 A549细胞株 2 4 h的凋亡诱导率为 :2 2 .80 %及 30 .1 5% ,48h的凋亡诱导率为 :41 .75%及 60 .0 4 % ,72 h的凋亡诱导率为 :56.38%及 73.30 % ,均分别高于相应时间点的 5- FU的作用 ,光镜下观察可见凋亡细胞体积缩小变圆 ,TUNEL染色后在免疫荧光显微镜下观察可见凋亡的细胞呈荧光染色阳性 ;结论 As2 O3 在体外可明显有效地诱导

Objective To study whether arsenic trioxide (AS 2O 3)will induce apoptosis of lung cancer cells.Methods Apoptosis rate and morphologic changes of A549 cells induced by As 2O 3 were observed under the microscope?fluorescence microscope. Results apoptosis rate induced by As 2O 3 of 1?2.5μmol/L was 22.80%,30.15% at 24h;41.75%,60.04% at 48 h and 56.38%,73.70% at 72 h,respectively,they were all higher than those acted by 5 Fu.Typical apoptotic cells were seen that the bodies of the cells became smaller and rounder than those before.Apoptotic cells were positive in fluorescence under fluorescence microscope.Conclusion As 2O 3 can induce apoptosis in lung cancer cells line A549.
