碱性成纤维细胞生长因子在门静脉高压症大鼠断流术后胃壁中的表达研究

STUDY OF THE EXPRESSION OF BASIC FIBROBLAST GROWTH FACTOR IN STOMACH AFTER DEVASCULARIZATION IN PORTAL HYPERTENSIVE RATS

目的 :探讨碱性成纤维细胞生长因子 (b FGF)在大鼠门静脉高压症断流术后胃底组织中的表达和意义。方法 :门静脉部分缩窄制备肝前型门静脉高压症 (PHPH)大鼠模型 ,并设立假手术对照 ;在术后 3周施行联合断流术 ,分别在断流术前及术后 1、3、5、7、14、30、90天取材 ;免疫组化检测 b FGF、第八因子相关抗原 (FV Ag)在胃底组织中的表达 ;对 FV Ag阳性血管进行微血管密度计数 (MVD) ,对 b FGF染色结果进行计算机图象定量分析。结果 :(1) MVD门脉高压组大鼠在断流术前较对照组显著增加 (P<0 .0 5 ) ,术后较术前明显下降 (P<0 .0 5 ) ,术后 14天明显高于术前 (P<0 .0 5 ) ,术后 3个月较术前升高近一倍 ;对照组断流术后 MVD逐渐增加 ,第 7天达到高峰 (P<0 .0 5 )。(2 ) b FGF门脉高压组在断流术前及术后 1天、3天、14天及 1个月均明显高于对照组 (P<0 .0 5 ) ,表达高峰在术后 3天和 14天 ,术后 1月仍显著高于术前水平 (P<0 .0 5 )。对照组在断流术后 3天 b FGF表达较术前明显升高 (P<0 .0 5 ) ,术后第 5天达高峰。结论 :b FGF对于胃壁中微血管的增生有重要作用。而 b

Objective:To study the expression and its significance of basic fibroblast growth factor (bFGF) in gastric fundus after devascularization in portal hypertensive rats.Methods:Pre-hepatic portal hypertensive (PHPH) rats were induced by partial portal vein stenosis,and the sham-operated (SO) rats were built simultaneously.Each of the two groups was divided into eight subgroups and sacrificed in pre-operation and 1,3,5,7,14,30,90 days after combined-devascularization respectively.We detected the expression of bFGF and factor VⅢ related antigen (FVⅢAg) in gastric fundus with the method of immunohitochemistry,envision-two steps,counted the microvessel density (MVD) through the results of FVⅢAg staining,and analysised the results of bFGF staining with the computed pathology image analysis system.Results:Compared with the SO rats,the MVD of PHPH rats increased significantly before devascularization (P<0.05),then decreased immediately after the operation (P<0.05) ,and increased gradually from the fifth day.Until 3 months after devascularization,it is almost twice than pre-operation stage (P<0.05).As to the SO rats the MVD increased step by step after operation,reached its peak in the seventh day. The expression of bFGF before operation and 1,3,14 days,1month after devascularization in PHPH rats was significantly higher than those in SO rats (P<0.05),and there were two pinnacles of bFGF experssion after operation:the third day and the fourteenth day respectively.Until one month after devascularization,the experssion level of bFGF in PHPH rats was still higher than preoperation stage (P<0.05), The expression of bFGF in So rats increased significantly on the third day after operation (P<0.05),reached its peak in the fifth day.Conclusion:Increased expression of bFGF promoted by combined-deascularization accounts for the increasing of MVD in gastric fundus.The increasing expression of bFGF may act as a promoter of the process.
