摘要
目的探讨 1d和 2d尾吊对小鼠胸腺细胞凋亡的影响。方法采用尾部悬吊小鼠模型模拟失重 ,碘化丙啶 (PI)染色法检测凋亡细胞染色体断裂 ,Annexin V分析法检测凋亡细胞膜的变化。结果尾吊小鼠未出现明显的染色体断裂 ;悬吊 1d ,总凋亡细胞数和早期凋亡细胞数呈增加趋势 ;悬吊 2d早期凋亡细胞数明显升高 ,总凋亡细胞数极显著增加。
Objective To investigate the effect of simulated weightlessnes s on apoptosis of thymus cells in mice. Method Tail-suspended mice w ere used as an animal model of simulated weightlessness; chromosome breakage wa s measured by PI staining,and changes of cell membrane were determined by Annexi n-V analysis. Result As compared with the control, there was no o bvious chromosome breakage; the total number of apoptotic cells and number of ea rly apoptotic cells increased in 1 d suspended group; the number of early apopto tic cells increased significantly and the total number of apoptotic cells increa sed very significantly in 2d suspended groups. Conclusion These resu lts suggested that simulated weightlessness could promote apoptosis of thymus ce lls.
