三七皂甙对烫伤小鼠巨噬细胞产生NO和TNF的作用 被引量:16

摘要 目的 研究三七皂甙对烧伤小鼠巨噬细胞 (MΦ)的NO产生和TNF分泌的调理作用。方法  4 50只昆明种小白鼠分成对照组 ( 1 8只 )、单烫组 ( 1 0 8只 )和治疗组 ( 32 4只 )。治疗组腹腔注射三七皂甙液 ,并分为大剂量组 (三七皂甙液 1 0 0mg/ 4h)、中剂量组 ( 50mg/ 4h)和小剂量组 ( 2 5mg/ 4h)。单烫组和治疗组分 6、1 4、2 6、50、74、98h观察时相点。用Griess反应法检测MΦ的NO产生量 ,以生物活性法测定MΦ的TNF分泌量。结果 三七皂甙能使烫伤小鼠MΦ的NO产生量从 ( 96 .2± 1 3.1 )nmol/ 1 0 6MΦ降至 ( 2 2 .1± 6 .2 )nmol/ 1 0 6MΦ、MΦ分泌TNF的量降低 55%。结论 三七皂甙对降低烧伤小鼠MΦ产生NO和MΦ分泌TNF均有显著的作用。 Objective To explore the effects of Panax pseudoginseng saponins (PS) on the nitric oxide production and TNF secretion by macrophages. Methods A total of 450 mice were divided into control (18), scalded (108) and treated (324) groups. PS were given intraperitoneally with differet doses, nomely large dose 100 mg/4 h, moderate dose 50 mg/4 h and small dose 25 mg/4 h. Production of NO by HΦ was determined with Griess reaction technique and secretion of TNF by MΦ with biological activity technique at different time points. Results Compared with scalded control, PS could decrease the amount of nitric oxide from (96.2±13.1) nmol/10 6MΦ to (22.1±6.2) nmol/10 6MΦ,and TNF to 55% of the control. Conclusion Panax pseudoginseng saponins might play important role in modulating the amount of nitric oxide and TNF produced by macrophage.
