摘要
目的 观察IGF I和TGF β1对幼年及成年兔关节软骨细胞增殖和代谢的作用。方法 采用体外培养兔关节软骨细胞的方法 ,利用 3H TdR和 35 S Na2 SO4掺入分别反映软骨细胞增殖和基质蛋白多糖的合成 ,应用TUNEL法检测软骨细胞的凋亡。结果 随年龄增加 ,关节软骨细胞凋亡增加 ,同时增殖和蛋白多糖的合成能力下降 ,而且对IGF I和TGF β1的反应性亦呈年龄相关下降。但是IGF I和TGF β1均可能在一定程度上以剂量依赖方式促进软骨细胞 3H TdR和 35 S Na2 SO4掺入增加。结论 外源性IGF I和TGF β1在体外可促进不同年龄兔关节软骨细胞增殖和基质合成 ,提示外源性生长因子在骨关节炎的防治上可能有一定意义。
Objective TO observe the effects of IGF Ⅰand TGF β 1 on the proliferation and matrix metabolism of chondrocytes in young and adult rabbit articular cartilage. Methods Chondrocytes were isolated from 3-12 months' old rabbit articular cartilage and cultured. Proteoglycan synthesis was measured by using 35 S Na 2SO 4 incorporation while proliferation was measured by 3H thymidine incorporation. The apoptotic chondrocytes were detected by using the TUNEL staining technique. Results With increasing of age, the TUNEL positive chondrocyte ratio increased whereas the chondrocyte proliferative and synthetic capacity decreased. However,IGF Ⅰ (3-30 ng/ml) and TGF β 1 (3-30 ng/ml) caused dose dependent increases in 3H thymidine and 35 S Na 2SO 4 incorporation.Conclusion IGF Ⅰ and TGF β 1 can promote, to some extent, chondrocyte proliferation and proteoglycan synthesis of different ages in vitro .Exogenous growth factors might be available in the treatment or prevention of osteoarthritis.
