摘要
目的 研究以NatipideⅡ空白脂质体包封低分子肝素形成脂质体的性质 ,为其制剂的开发提供参考。方法 以超滤法测定脂质体包封率和不同温度下的渗漏率 ,以透射电镜观察其形态 ,以表面电位粒径仪测定脂质体颗粒表面电荷及zeta电位。结果 表观重量包封率 43 .18% ,颗粒为圆形或椭圆形 ,平均粒径为 2 0 0nm ,平均zeta电位为 - 6 6 .45mV ,在 4,15 ,2 5℃放置 3个月后渗漏率分别为 10 .2 6 % ,16 .5 8%和 2 5 .5 4%。结论 NatipideⅡ空白脂质体对低分子肝素具有较强的包封能力 ,形成的脂质体较为稳定 。
OBJECTIVE: To study the characteristics of liposomes of LMWH encapsulated by proliposomes. METHODS: Encapsulated ratio of liposomes and leaked ratio at different temperature were determined by ultra-filtration, the shape of liposomes was observed under the electronic microscope and zeta potential of liposomal vesicle was determined with the instrument. RESULTS: The apparent weight encapsulated ratio of liposomes was 43.18%. The vesicles were globular or elliptic. The mean diameter was about 200 nm. The mean zeta potential was -66.45 mV, and the leaked ratio was respectively 10.26%, 16.58%, and 25.54% at 4, 15, 25°C after 3 months. CONCLUSION: The proliposomes had well encapsulating ability for LMWH. The liposome system of LMWH was stable at low temperature and worthy further exploiting.
