海藻酸盐-多聚赖氨酸-海藻酸盐微胶囊膜的强度和生物相容度测定 被引量:19

摘要 目的 :了解我们所制作的海藻酸盐 多聚赖氨酸 海藻酸盐 (APA)微囊的生物相容性、机械和化学强度。方法 :用静电微囊发生器制作海藻酸盐 多聚赖氨酸 海藻酸盐的APA微胶珠及枸橼酸钠液化微珠芯的APA微胶囊。用振摇法比较微囊与微珠的机械强度。用不同酸碱度溶液测定微囊的化学强度。用微囊小鼠腹腔移植法观察APA微囊的生物相容度。结果 :①机械振摇 48h的破损率 :微囊为 5 % ;微珠为 31% ;②Ph 7 4对APA微囊的化学损伤最小 ;③移植后 1周、2周、4周、8周和 10周从小鼠腹腔内取出的微囊 95 %~ 98%为完整、圆形、表面光滑、无明显的结缔组织包绕。结论 :我们所制作的APA微囊具有较好的生物相容度。 Objective:To know the biocompatibility of APA microcapsules we made and compare the difference in the mechanical strength of two kind of capsules.Methods:The alginate poly L lysine alginate (APA) microcapsules were made by the static generator The microcapsules were divided into typeⅠor typeⅡ according to using sodium citron to liquefy bead core or not The mechanical strength of the microcapsules was measured by shaking method The chemical intensity of APA microcapsules was detected by using vary acerbity and alkalinity liquor The biocompatibility of APA microcapsules was observed by transplanting them into intraperitoneum of mice.Results:The broken ratio of 48h mechanism shaking: type Ⅰ5% and type Ⅱ31%,which indicated that type Ⅰhad better mechanical strength than type Ⅱ had 2 pH7 4 were suitable for the microcapsules 3 The microcapsules were taken out from mice intraperitoneum after transplanted 1、2、4、8 and 10 weeks, and 95 98 percent of these capsules were of integrity, rotundity, surface smooth, without obviously bundled by connective tissue. Conclusion:The type Ⅰ APA microcapsules we made had good biocompatibility、mechanical strength and chemical intensity.
