摘要
目的 探讨脑缺血再灌注模型大鼠血脑屏障是否对高分子量的多肽 -碱性成纤维细胞生长因子 (b FGF)通透。方法 采用大脑中动脉腔内阻塞模型 ,股静脉滴注 1 2 5I- b FGF,对大脑冠状冰冻切片进行放射自显影 ,图象分析测定光密度。结果 正常对照大鼠脑冠状切片放射自显影结果显示脑轮廓影 ,且为全脑均匀分布 ;模型鼠除了显示脑轮廓影外 ,可见缺血病灶侧影像较健侧及正常鼠浓集。光密度测定结果显示模型鼠病灶侧与其健侧及正常鼠同侧的光密度值差异显著 (t=3.54,P<0 .0 5;F=39.6,P<0 .0 5)。结论 1 2 5I- b FGF经股静脉滴注后 。
? Objective To investigate whether bFGF,a high molecular weight polypeptide,could cross blood brain barriers to enter ischemic areas in a model of focal cerebral ischemia and reperfusion in rats.Methods A model of middle cerebral artery occlusion and reperfusion was established by intraluminal suture in mature SD rats. 125 I labeled bFGF was infused intravenously.Labeled bFGF in cerebral tissues was displayed by autoradiography and optical density was determinated.Results Autoradiography and determination of OD showed that OD was 0 255 1±0 021 3 in the ischemic and 0 212 0±0 017 2 in the normal areas of model rats( P <0 05).and 0 210 2±0 016 2 in the same sides of normal rats( P <0 05).Conclusions Following intravenous administration of 125 I labeled,bFGF,labeled bFGF could cross blood brain barriers to enter ischemic areas in a model of focal cerebral ischemia and reperfusion in rats.
