摘要
目的 研究五子衍宗丸及其拆方对老年大鼠骨骼肌线粒体 DNA缺失、线粒体呼吸链酶复合体及 ATP合成的影响。方法 用聚合酶链反应 (PCR)、酶动力学和生物发光技术进行检测。结果 老年大鼠骨骼肌线粒体 DNA缺失较青幼年大鼠明显增多 ,线粒体呼吸链复合酶 活力及ATP合成量较青年大鼠明显减少。五子衍宗丸及其拆方的枸杞子、菟丝子可减少老年大鼠骨骼肌线粒体 DNA缺失 (P<0 .0 1 ) ,提高线粒体呼吸链复合酶 活力 (P<0 .0 5) ;五子衍宗丸还可提高老年大鼠骨骼肌线粒体 ATP的合成 (P<0 .0 5)。结论 五子衍宗丸、枸杞子和菟丝子对老年大鼠线粒体
? Objective To investigate the effect of Wuzi Yangzong pill and its disassembled prescription on skeletal muscle mitochondrial DNA(mtDNA) deletion,respiratory chain complexes and ATP synthesis in aged rats.Methods By using PCR,enzyme kinetics and bioluminescence technique.Results The deletion of skeletal muscle mtDNA increased significantly in 24 month old rats compared with 1 and 6 month old rats,and the activities of mitochondrial respiratory chain complexes Ⅳ and ATP synthesis decreased significantly in 24 month old rats compared with 6 month old rats ( P <0 05,0 01).Wuzi Yanzong pill and its disassembled prescriptin of Fructus Lycii(wolfberry Fruit)and Semen Cusuctae(Dodder seed) could reduce the mitochondrial DNA deletions and raise the ativities of mitochondrial respiratory chain complexes Ⅳ in aged rats( P <0 05,0 01),Wuzi Yanzong pill could also raise the mitochondrial ATP synthesis in aged rat skeletal muscle( P <0 05).Conclusions Wuzi Yanzong pill,Fructus lycii and Semen Cusuctae have protective effect on oxidative damage of skeletal muscle mitochondrial DNA in aged rats.
