Determination of tetrodotoxin by RP-IP-HPLC

目的 :建立了反相离子对 -高效液相色谱法测定河豚毒素含量的方法。方法 :选用 SHIMADZU ODS色谱柱 (15 0× 6 mm5 μm) ,以 0 .2 % (v/v)醋酸液为流动相 ,流速为 1.2 m L/m in,检测波长为 2 30 nm。结果 :河豚毒素线性范围 2 0～ 10 0 μg/m L,r= 0 .996 0 (n=5 ) ;回收率为 93.32 % ,RSD=5 .41% (n=5 ) ;日内精密度为 6 .10 % ,日间精密度为 7.42 % (n=5 ) :最低检测限5 0 ng。结论 :方法准确、快速、简便 。

OBJECTIVE:A RP IP HPLC method was used to determine the content of tetrodotoxin. METHOD: The HPLC was performed on a SHMADZU ODS Column (150×6 mm 5 μm) with 0.2% (v/v) acetate solution as the mobile phase(flow rate 1.2mL/min) and the detection wavelength was at 230nm. RESULTS: The linear rang of tetrodotoxin was 20～100 μ g/mL(r=0.9960,n=5).The mean recovery was 93.32%,RSD=5.41%(n=5).The variation of within day was 6.10% and that of day to day was 7.42%(n=5).The limit of detection was 50ng. CONCLUSION: The method is simple,sensitive and accurate,and can be applied to the rapid determination of tetrodotoxin.
