中国人胶质细胞源神经营养因子基因的克隆及测序

摘要 目的 为了克隆中国人胶质细胞源神经营养因子(GDNF)基因,从而为开发重组人GDNF治疗神经变性疾病打下基础。方法 根据从GeneBank中调出的人GDNF基因全序列设计引物,以中国人外周血白细胞提取的基因组DNA为模板,通过PCR扩增出中国人GDNF基因片段,克隆到pGEM-T载体上,进行序列分析。结果 我们得到的一种425bp DNA片段。经与GeneBank中已注册的人GDNF基因比较,序列完全相同。结论 正确的中国人的GDNF基因已克隆成功。 Objective To clone Chinese GDNF gene in order to develop recombinant human GDNF for treating the neurodegenerate diseases. Methods Based on the complete DNA sequence of human glial cell line-derived neurotrophic factor (GDNF) gene from GeneBank, a pair of primers were designed and PCR was carried on template prepared as Chinese genome DNA, which was then cloned into pGEM-T vector and sequenced. Results A DNA fragment of 425bp was obtained. The nuc-leotide sequences are the same as those of human GDNF gene already published. Conclusion The correct Chinese glial cells line-derived GDNF gene was obtained.
