摘要
目的 研究大多功能蛋白酶 (LMP) 7基因与 1型糖尿病 (DM )易感性的关系。方法采用聚合酶链反应 限制性片段长度多态性技术对 71例 1型DM患者及 86例正常对照进行LMP7基因分型。根据发病年龄将 1型DM患者分为 3组 ,A组≤ 14岁 ,B组 15~ 30岁 ,C组≥ 31岁。结果 1型DM组LMP7 B/B频率较对照组显著降低 (39%vs 5 8% ,P <0 .0 5 ) ,LMP7 B/A频率显著升高 (5 4%vs 31% ,P <0 .0 1)。LMP7各基因型及等位基因频率在糖尿病各发病年龄组间差异无显著性。结论 LMP B/B可能是 1型DM的保护基因型 ,携带该基因型的人群患者 1型DM危险性降低。LMP7 B/A可能是 1型DM的易感基因型 ,携带该基因型的人群患 1型DM危险性增加。LMP7基因与
Objective To study the relationship between large multifunctional proteasome (LMP) 7 gene polymorphism and susceptibility of type 1 diabetes mellitus (DM). Methods The genotyping of LMP7 gene was determined by polymerase chain reaction restriction fragment length polymorphism (PCR RFLP) in 71 type 1 DM patients and 86 healthy persons (as controls). Furthermore, the type 1 DM patients were divided into 3 groups according to the age of diabetic onset. Group A was ≤14 years, group B 15~30 years, group C≥31 years.Results The frequency of LMP7 B/B was decreased significantly (39% vs 58%, P<0.05), and that of LMP7 B/A increased significantly (54% vs 31%, P<0.01) in type 1 DM patients. Both LMP7 genotypes and alleles of LMP7 gene did not show differences among various age groups of diabetic onset. Conclusions The LMP7 B/B may be a protective genotype, and the persons with LMP7 B/B may have a decreased risk of suffering from type 1 DM. The LMP7 B/A may be a susceptible genotype, the persons with LMP7 B/A may have an increased risk of type 1 DM. The LMP7 may have no association with the age of diabetic onset.
