摘要
以多孔聚酯材料作固定化载体,考察了载体吸附固定化黑曲霉菌丝细胞的条件,当菌丝体细胞与载体预培养的条件为pH值5.0、孢子浓度为10~5个/ml、固液比为1/75时,有利于菌丝体的生长、吸附固定及发酵产酶。在产糖化酶的发酵过程中,与游离菌丝体细胞相比,发酵过程持续产酶时间有一定程度的延长,产糖化酶活力始终高于游离菌丝体。
The conditions of immobilization of the mycelia of Aspergillus niger to be absorbed within the Porous polyurethane supporter were investigated and, the aspect of its glucoamylase fermentation of immobilized and freer cell were compared too. By reducing the immobilized viabe spore loading(to 10~5spores ml^(-1) and ad- justing the pH value(at the range of 5. 0 to 5. 5) as well as the solid/liquid ratio(at 1/75) in liquid culture, absorption and immobilization of mycelia were well promoted and, an increase in glucoamylase activity was given rise to in media. By immobilization, the duration of glucoamylase producing was extended and cell growth was inhibited to some extent when compared to free cell in fermentative process.
