摘要
目的 : 研究β-胡萝卜素在离体心肌细胞缺氧损伤时的保护作用。方法 : 建立幼鼠心肌细胞缺氧损伤模型 ,观察乳酸脱氢酶 ( LDH)、氧自由基 ( OFR)、超氧歧化酶 ( SOD)、丙二醛( MDA)等项过氧化物造成心肌损伤的检测指标 ,并对经 β-胡萝卜素预处理的心肌细胞缺氧模型进行观察。结果 : 乳鼠心肌细胞经 2 h缺氧损伤后 LDH,MDA,OFR,SOD等项指标均与对照有显著性差异 ( P<0 .0 1 )。用 β-胡萝卜素预处理的心肌细胞缺氧后经检测其保护作用随浓度梯度增强。结论 : 1 0 -2和 1 0 -1mmol/L的 β-胡萝卜素对心肌细胞缺氧具有抗氧化保护作用。
Objective: To study the protective effect of β carotene in anoxia injured cardiomyocytes. [WT5FZ]Methods: The cultured rat neonatal cardiomyocytes were subjected to 2h anoxia and the changes of lactate dehydrogenase(LDH) activity, malondialdehyde (MDA), oxygen free radical (OFR) contents and superoxide dismutase (SOD) activity were detected. The effects of pre treatment with β carotene were then investigated. Results: The LDH, MDA, OFR and SOD of cardiomycytes were changed significantly after 2h anoxia (P<0.01).However, the β carotene pre treated cardiomyocytes showed less changes. Conclusion: β carotene (10 -2 ~10 -1 ) mmol/L can protect cardiomyocytes from anoxia injury.
