摘要
目的 研究稳恒磁场对肢芽细胞增殖和分化的强度效应关系。方法 利用大鼠胚胎肢芽细胞进行原代培养 ,观察培养物经不同强度的稳恒磁场 ,对细胞增殖和分化的影响 ,并利用图像分析细胞形态的变化。结果 各组磁场强度能明显抑制肢芽细胞增殖和分化 ,集落形成率明显减少 ,并显示强度效应关系。拟合细胞分化和增殖抑制曲线 ,计算得大鼠半数分化抑制强度 (ICD50 )为 35mT ,半数存活抑制强度 (IVD50 )为 48mT。结论 抑制细胞分化、增殖可能是稳恒磁场强度致发育危害的结果。
Objective To explore the effect of static magnetic field (SMF) on differentiation and proliferation of rat embryo limb bud cell.Methods The micromass culture of embryo limb bud cell was applied to study the effect of varieties of SMF (10 mT, 20 mT, 30 mT, 40 mT, 50 mT, 60 mT).Results 14 days embryo limb bud cell of rat was subjected to culture for 5 days. The results showed that SMF could inhibit differentiation of the cell without affecting cells proliferation. The concentration of 50% inhibition of cell differentiation (ICD 50 ) was 35 mT. The concentration of 50% inhibition of cell proliferation (IVD 50 ) was 48 mT. Conclusion The static magnetic field might result in abnormality of the cell in embryogensis.
