摘要 目的 探讨纤维连接蛋白、 型胶原蛋白对晶状体上皮细胞粘附的影响 ,研究Arg-Gly-Asp-Ser( RGDS)肽抑制晶状体上皮细胞与基质蛋白粘附的作用。方法 应用MTT法观测纤维连接蛋白、 型胶原蛋白对体外培养的牛晶状体上皮细胞的粘附作用 ,其次测定 RGDS肽对牛晶状体上皮细胞与细胞外基质蛋白粘附和细胞增殖的影响。结果 牛晶状体上皮细胞能够粘附到包被有不同浓度的纤维连接蛋白、 型胶原蛋白培养板表面上 ,呈现浓度效应特点 ;RGDS肽能够抑制牛晶状体上皮细胞在基质蛋白上的粘附。结论 纤维连接蛋白、 型胶原蛋白能够介导晶状体上皮细胞粘附到后囊上 ,在后囊混浊发生过程中起到重要的促进作用 ;在细胞水平上证实了 RGDS肽通过竞争细胞外基质蛋白与晶状体上皮细胞表面的整合素受体结合 ,阻止细胞粘附和增殖 ,具有潜在的防治后囊混浊的作用。 Objective To probe into the influence of fibronectin and typeⅠcollagen on promotion of lens epithelial cells(LECs) adhesion,and to investigate the role of Arg Gly Asp Ser(RGDS) peptide in the adhesion between LECs and matrix proteins.Method MTT assay was applied to observe adhesion of bovine LECs cultured in response of fibronectin , typeⅠcollagen in vitro, and to determine the influence of RGDS peptide on adhesion and proliferation of LECs. Result Bovine LECs adhered to surfaces which were coated with varying concentration of fibronectin and typeⅠcollagen, and the adhesion of the cells was dependent on the concentration of the coated substance. RGDS peptide inhibited the adhesion between bovine LECs and fibronectin/typeⅠcollagen. Conclusion Fibronectin and typeⅠcollagen mediate the attachment between posterior capsule and LECs, which may play a promotive role in the occurrence of posterior capsular opacification(PCO). RGDS peptide inhibits cellular adhesion and proliferation by competing with matrix protein for binding to the integrin receptors of LECs surfaces, and then is potentially useful for preventing PCO.
