摘要
目的 检测生长因子抑制剂苏拉明对体外原代培养的牛晶状体上皮细胞 (bovinelens epithelial cells,BL ECs)的抑制作用。方法 原代培养 BL ECs,在其生长的第 3代时分别加入含有苏拉明 0、30、10 0、2 0 0、30 0、5 0 0 mg· L- 1的 DMEM培养液 ,分别在其作用后的4h、2 d、4d、6 d用 MTT染色 -比色法检测细胞的生长情况 ;在原代培养的第 3代细胞中分别加入含 15 0 m g· L- 1 和 35 0 m g· L- 1 苏拉明的 DMEM培养液 ,分别在其作用后的 6 d和3d将细胞消化 ,进行流式细胞仪检测细胞的增殖周期。结果 浓度为 30 mg· L- 1和 30 0 mg· L- 1 的苏拉明分别作用于 BL ECs 4d或 4h,便产生了明显的抗 BL ECs增殖作用 (分别为P <0 .0 1和 P <0 .0 5 ) ,其抑制程度随作用时间的延长和试剂浓度的升高而逐渐增强。苏拉明将 BL EC阻抑于细胞增殖周期的 G2 / M期。结论 苏拉明有效抑制了体外原代培养的 BL ECs的生长 ,如进一步进行在体研究 ,可望成为防治白内障术后后囊膜混浊的一种良药。
Objective To study the preventing effect of suramin on lens epithelial cells (LECs) in vitro.Methods Bovine lens epithelial cells (BLECs) were cultured in primary manner. DMEM with six different doses of suramin (0, 30, 100, 200, 300 and 500mg·L -1 ) were added to the cultured BLECs respectively during its third growth generation. The growth of the BLECs were examined by MTT method at its four different time (4h, 2d, 4d and 6d respectively); DMEM with 350mg·L -1 and 150mg·L -1 suramin were added to cultured BLECs respectively. When the BLECs had grown for 3d and 6d, they were collected and their growth cycle was checked with lapse cytometry.Results When suramin with its doses≥30mg·L -1 and≥300mg·L -1 for 4d or 4h respectively, BLECs were inhibited significantly (P<0.01 and P<0.05 respectively). The preventing effect showed dose dependent and time dependent manner. BLECs might be blocked or head a prolonged G 2 and/or M phase in their cell cycle in suramin treated group.Conclusion Suramin can effectively inhibit BLECs' growth in vitro. If it is used in vivo, suramin will become a good drug to prevent PCO.
