摘要 应用RT -PCR技术 ,从大腹圆蛛 (Araneusventricosus)壶腹腺中扩增出鞭毛样丝蛋白基因 (flagelli formsilkproteingene) ,经 1 5 %琼脂糖凝胶电泳分离 ,WizardPCRPrepsDNAPurificationSystem回收后 ,将其克隆在pGEM -T载体中 ,经限制性核酸内切酶鉴定和核苷酸序列分析证实 ,构建的重组质粒pSF1中含有蜘蛛鞭毛样丝蛋白基因 ,且含有 The flagelliform silk protein gene was amplified from ampullate of Araneus ventricosus by RT-PCR,isolated by 1.5% agarose gel electrophoresis, recovered by Wizard PCR Pres DNA purification system, and then inserted into a clone vector pGEM-T by cohesive end ligation. The recombinant plasmid pSF1 was studied indetail by restriction endonuclease analysis and nucleotide sequencing. The results showed that the recombinant plasmid pSF1 contains flagelliform silk protein gene with three repetive sequences.
