摘要
目的 :探讨白细胞介素 10 (interleukin 10 ,IL 10 )对血管活性肽尾加压素Ⅱ (urotensinⅡ ,UⅡ )诱导的大鼠血管平滑肌细胞 (vascularsmoothmusclecell,VSMC)增殖的影响及其作用机制。方法 :在培养的大鼠主动脉VSMC上 ,采用3H 胸腺嘧啶 (3H TdR)参入测定VSMCDNA合成速率 ,Westernblot及免疫沉淀法测定粘着斑激酶(focaladhensionkinase ,FAK)的表达及活性。结果 :IL 10 (10 -10 ~ 10 -8g·ml-1)呈浓度依赖的方式抑制UⅡ (10 -8mol·L-1)诱导的VSMC3H TdR参入增加 (P <0 .0 5或P <0 .0 1)和FAK的活性上调 (P <0 .0 5 ) ,但各组间FAK的蛋白表达差异无显著性。结论 :IL 10可能通过抑制FAK活性的上调 。
Objective: Exploring the effect of interleukin 10 (IL 10) on the changes of urotensin Ⅱ (UⅡ) induced rat vascular smooth muscle cell (VSMC) proliferation and focal adhesion kinase (FAK) signal. Methods: On cultured VSMC of rat, 3H TdR incorporation was used to evaluate the speed of DNA synthesis, Western blot and immunoprecipitation were applied to assay the expression and activity of FAK. Results: IL 10(10 -8 -10 -10 g·ml -1 )inhibited the increasing of 3H TdR incorporation and FAK activity, which was induced by UⅡ( P <0.05 or P <0.01). But there was no difference of the expression of FAK among the groups. Conclusion: IL 10 can inhibit the VSMC proliferation by downloading the activity of FAK, which stimulated by UⅡ.
