胶质瘤细胞ING1基因表达水平与肿瘤细胞增殖和凋亡的关系研究 被引量:3

摘要 目的 探讨胶质瘤细胞ING1基因表达水平及其与肿瘤细胞增殖活性和凋亡程度的关系。方法 采用原位杂交及免疫组织化学染色方法 ,分别检测 71例不同级别的人胶质瘤组织ING1mRNA及ING 1、PCNA蛋白 ,并采用TUNEL法检测原位细胞凋亡的情况。结果 ING 1mRNA的阳性表达率为 94.6% (3 5 / 3 7) ,ING 1蛋白的阳性表达率为 90 .1% (64 / 71) ,两者的表达水平呈正相关 (γ =0 .714 ,P <0 .0 1) ,均随肿瘤恶性程度的增高而降低 ,具有显著性差异 (P <0 .0 5 )。ING 1表达水平降低时伴随着凋亡程度的降低及增殖活性的增强。结论 胶质瘤细胞中ING1基因的表达水平与肿瘤恶性程度密切相关 ,并随肿瘤恶性程度的增加而相应降低。其表达异常的改变主要是转录水平调控异常的结果 ,并可能通过促进细胞增殖和抑制细胞凋亡 ,在胶质瘤的发生、发展中发挥重要作用。 Objective To investigate the expression level of ING1 gene and their effects on cell proliferation activity and apoptosis degree in human gliomas.Methods The expression of ING1 mRNA,ING1 protein,proliferating cell antigen(PCNA) and apoptosis in 71 human glioma specimens with different malignant grades were studied by immunohistochemistry in situ hybridization and in situ cell death detection(TUNEL) methods.Results The expression rate of ING1 mRNA and ING1 protein were 94.6%(35/37) and 90.1%(64/71),respectively.They were correlated positively with each other(γ s=0.714,P<0.01),and they decreased as increase of the malignant grade (P<0.05).PCNA activity was increased with decline of the ING1 gene expression,while apoptosis degree decreased.Conclusion The expression levels of ING1 gene are correlated closely with the degree of tumor malignancy in glioma cells.They decrease with the increase of tumor grade.The abnormal decrease of ING1 gene expression might be an important factor in the genesis and development of gliomas.Probably ING1 exerts an important function in the genesis and development of gliomas by promoting cell proliferation and suppressing cell apoptosis.
