Rb、p21、MMP-2和LN-R在食管鳞癌中的表达及其与侵袭和转移的关系

Relation of Rb,p21,MMP-2 and LN-R Protein Expression to Invasion,and Metastasis in Esophageal Squamous Cell Carcinoma

目的 探讨Rb、p2 1、MMP 2和LN R在食管鳞癌中的表达及其与侵袭和转移的关系。方法 应用免疫组化S P法检测Rb、p2 1、MMP 2和LN R在食管鳞癌、癌旁、肌层浸润灶和淋巴结转移组织中的表达。 结果 p2 1、MMP 2和LN R在癌组织中的表达高于癌旁组织 ,分化Ⅲ级的高于Ⅰ、Ⅱ级 ,有淋巴结转移组高于无淋巴结转移组 ,具有非常显著性差异 (P <0 .0 1)。而p2 1、MMP 2的表达与肿瘤的浸润深度有明显相关性 (P <0 .0 5 )。Rb的表达与肿瘤的浸润深度、分化程度和淋巴结转移呈负相关(分别为P <0 .0 5、P <0 .0 1和P <0 .0 5 )。结论 抑癌基因Rb的失表达和 p2 1、MMP 2及LN R蛋白的过表达与食管鳞癌的分化程度、浸润和转移关系密切。检测Rb、p2 1、MMP 2和LN R有助于对食管鳞癌的生物学行为及预后的评价。

Objective To investigate the relation of Rb、p21、MMP-2 and LN-R protein expression to invasion and lymph node metastasis of esophageal squamous cell carcinoma.Methods The expression of Rb,p21 MMP-2 and LN-R protein was examined by immunohistochemieal S-P method in tumors tissues,adjacent tissues and lymph node metastasis.Results The frequency of p21,MMP-2,LN-R protein expression of tumor tissues was remarkably higher than that in adjacent tissues,It was remarkably higher in differentiated grading Ⅲ than that in differentiated grading Ⅰ、Ⅱ;and remarkably higher in lymph node metastasis group than that in non-metastasis group(P<0.01).The expression of p21,MMP-2 was significantly correlated with infiltrating depth of tumor(P<0.05).The expression of Rb protein in esophageal squamous carcinoma was inversely related to the depth of invasion,differentiation degree and lymph node metastasis(P<0.05、P<0.01 and P<0.05 respectively).Conclusion It is suggested that loss of Rb,p21,MMP-2 and LN-R protein overexpression is related to differentiated,infiltrating and metastasis of esophageal squamous carcinoma.The examination of Rb,p21,MMP-2 and LN-R protein is helpful for evaluation of biological behaviour and prognosis in esophageal squamous carcinoma.
