摘要
目的 :探索一种用可见分光光度计分析马钱子中士的宁和马钱子碱的方法。方法 :使用薄层色谱法分离提纯马钱子的氨性氯仿 (氯仿 -氨水 =9∶ 1)提取液 ,于 42 5 nm波长处检测显色后的氯仿萃取液 ,同时又与用紫外分光光度法直接检测分离出来的士的宁 (2 5 4nm )和马钱子碱 (2 6 4nm )的方法进行了比较。结果 :4.0~ 16 .8μg· m l- 1 的士的宁和马钱子碱染色后具有良好的线性关系 (r士 =0 .9991,r马 =0 .9997)。回收率为 99.82 % ,RSD=0 .87。结论 :酸性染料比色法准确、灵敏、可以替代紫外分光光度法分析马钱子中的士的宁和马碱子碱 (P<0 .0 0 1)
OBJECTIVE:To find a method which can analysis strychnine and brucine in semen strychni using 722 or 721 type spectrophotometer.METHODS:Separating and purifying the extract from semen strychni by thin layer chromatography with the extractant of 9∶1 CHCl 3-aqNH 3 solu mixture. Examining the CHCl 3 solvent phase at 245nm extracted from the dyed aq solu of the alkloid .Using UV spectrophotometer,to examine the separated strychnine at 254nm, and brucine at 264nm.RESULTS: Strychnine or brucine has good linear relationship in 4.0~16.8 μg·ml -1 (r 1=0.999 1, r 2=0.999 7). The average added recovery of ADC was 99.82%,RSD=9.87%.CONCLUSION: TLC ADC is accurate and sensitive, which can examine and strychnine brucine in semen strychni in place of UV spectrophotometry(P<0.01).
