摘要
以高效液相色谱法作为血液中土霉素含量的检测方法,研究了单次静注土霉素(20mg/kg)后在正常和沙门氏菌内毒素引起发热猪体内的动力学行为。结果表明,土霉素在正常猪体内适合无吸收三室开放模型,而在发热猪体内适合无吸收二室开放模型,发热改变了土霉素在猪体内的动力学行为。在正常猪,其消除半衰期(t0.5β)和总体消除率(CLB)分别为6.40h和0.174L/kg·h,在发热猪,则分别为6.13h和0.109L/kg·b.以1μg/ml的土霉素血清浓度为最小有效浓度,求出的维持有效浓度时间为20.34h和26.42h。并根据求出的药动学参数,推荐了一个给药方案。
The pharmacokinetics of oxytetracycline (OTC), given as a single intravenous dose (20mg/kg·B·W), were investigated in normal and febrile swine. OTC conccnration in plasma was determined by HPLC. The pharmacokinetic behaviour of OTC was described by three—compartment open model with a half-life of 6.40h and two—compartment open model with a half—life of 6.13h in febrile swine. The pharmacokinetic behaviour of OTC was changed by fever. A proper dosage regimen based on this research was proposed in both.
