摘要
目的 :探讨L 精氨酸 /一氧化氮合酶途径在脂多糖 (LPS)对人肾系膜细胞增殖中的作用。方法 :利用四甲基偶氮唑 (MTT)法检测L 精氨酸对脂多糖刺激的人肾系膜细胞活力的影响 ;利用硝酸还原酶法测定细胞上清液中亚硝酸盐含量 ,推测L 精氨酸代谢产物一氧化氮 (NO)对LPS刺激的人肾系膜细胞增殖的影响。结果 :L 精氨酸抑制LPS刺激的人肾系膜细胞增殖 ;细胞上清液中亚硝酸盐含量无明显变化。结论 :LPS不能刺激人肾系膜细胞产生大量NO ,L 精氨酸对LPS刺激的人肾系膜细胞增殖的抑制作用与L 精氨酸 /一氧化氮合酶途径无关。
Objective To study the effect of L arginine/NOS pathway on LPS stimulated human renal mesangial cell proliferation.Methods MTT assay method was used to examine mesangial cell activity with L arginine stimulating,and nitric acid reducing enzyme assay was used to check the nitrite in supernate in order to indirectly speculate the effect of NO on LPS stimulated mesangial cells.Results L arginine inhibited the proliferation of LPS stimulated mesangial cells. There were no statistical difference of the quantities of nitrite in supernate among the groups.Conclusion LPS cannot cause human mesangial cells produce abundant NO,the inhibitive effect of L arginine on LPS stimulated human mesangial cell proliferation is independent of L arginine/NOS pathway.
