摘要
本研究在建立GMA诱导的HELF体外恶性转化模型的基础上 ,采用mRNA差异显示 (DD PCR)技术 ,对恶性转化细胞同正常细胞之间基因的差异表达状况进行了比较分析 ,共克隆到 18个差异表达cDNA片段。其中 ,17个DNA片段与已知基因高度同源 ,另外一个为新基因序列。Northern杂交证实 ,分别有 8个和 3个基因在GMA诱导的恶性转化细胞中表达增加或降低。结果提示 ,这些基因可能参与了GMA诱导的细胞恶性转化过程 。
On the basis of establishment of an in vitro malignant transformation model of human embryonic lung fibroblasts (HELF) induced by giycidyl methacrylate (GMA), gene expression of the transformed cells and of controls was compared by mRNA differential display (DD PCR) technique. Eighteen differentially expressed cDNA fragments were cloned and sequenced, of which 17 were highly homologous to known genes and one was novel. Results of Northern hybridization showed that expression of 8 genes was up regulated and 3 genes down regulated respectively. These results suggested that the above genes were involved in the malignant transformation induced by GMA, and the information provided clues to elucidate the acting mechanisms of the compound.
