T淋巴细胞表面的高亲和力白细胞介素 2受体 (IL 2R)由α ,β及γ三种亚基组成 ,其中只有IL 2Rα亚基具有特异结合IL 2的活性 ,目前对IL 2Rα亚基的确切的功能尚不清楚。我们将IL 2RαcDNA反意表达质粒转染Jurkat细胞后 ,经Western印迹 ECL分析发现 ,细胞内出现了细胞凋亡早期特异的多聚二磷酸腺核糖聚合酶 [Poly(ADP ribose)polymerase ,PARP]的 89kD断裂片段 ,此结果提示 ,IL
High affinity receptor for interleukin 2 (IL 2R) is composed of three subunits, the α, β and γ. Although IL 2Rα is the only subunit which specifically binds to IL 2, its cellular function remains unclear. In this paper, we used the Western ECL technique to explore the occurrence of a 89 kilo Dalton fragment of poly (ADP ribose) polymerase, PARP that usually represents an irreversible signal of apoptosis. We found that transfection of an expression plasmid of antisense IL 2Rα cDNA into Jurkat cells induces cleavage of PARP indicating that IL 2Rα subunit participates in the regulating apoptosis in Jurkat cells.
