摘要
应用体外甲基化酶M SssI处理和甲基化敏感单链构象分析法 (Methylation sensitivesingle strandconformationanalysis,MS SSCA )及DNA测序技术 ,研究正常人乳腺癌易感基因 1(breastcancer 1,BRCA1)启动子区 5’端CpG岛13个CG二核苷酸位点的甲基化模式。对甲基化酶处理时间不同得到的 8种有差异MS SSCA迁移带型进行测序 ,证实为 8种不同的甲基化模式。
The methylation patterns of 13 CG dinucleotide sites in CpG islands of BRCA1 promoter from normal blood DNA after treatment with methylase M SssI were analyzed by methylation sensitive single strand conformation analysis (MS SSCA) and DNA sequencing Eight different migration patterns observed by MS SSCA were confirmed as eight different methylation patterns by DNA sequencing These researches will provide reference for the study of BRCA1 gene methylation patterns in clinic breast cancer samples.
