大麦胚芽鞘切段伸长的适宜条件和对网斑病菌毒素的反应 被引量:1

摘要 大麦胚芽鞘切段在稀释10倍的柠檬酸-磷酸缓冲液中的伸长量显著大于原液(原液由K2HPO4·3H2O 2.351g+柠檬酸1.019g+蔗糖20g,加水定容至1,000mL配制而成,pH=5.0)中的伸长量,长为10~12mm的胚芽鞘顶端3mm下的5mm长切段在单位时间内的伸长量显著大于其后的各切段,胚芽鞘越长,芽鞘切段伸长越慢,5‰~40‰浓度的蔗糖均能促进芽鞘切段的伸长,蔗糖浓度为20‰时促进作用最明显。大麦网斑病菌无菌培养滤液对感病品种甘啤1号芽鞘切段伸长的抑制作用显著大于对抗病品种莫特44号的抑制作用。 :The barley coleoptile sections in vitro grew significantly faster in 10-fold dilution of buffer solution of citric acid-potassium phosphate than that in the original. The optimum buffer solution pH for coleoptile sections to elongate was 5.0.The first 5-mm-long sections below apical 3mm length of coleoptile elongated faster than following 5-mm-long sections. The longer the coleoptiles grew, the slower the coleoptile sections elongated.Sugcrose concentrations ranging from 5g/kg to 40g/kg promoted the elongation of coleoptile sections with the optimum concentration at 20g/kg. The growth inhibition of coleoptile sections in vitro increased with the rising of concentration of the mycotoxin produced by barley net-blotch disease pathogen (Pyrenophora teres). The growth of coleoptile sections of susceptible barley cultivar Ganpi-1-hao were inhibited more significantly than resistant cultivar Mote-44-hao by the mycotoxin.
