分蘖洋葱茎尖愈伤组织诱导及植株再生 被引量:22

摘要 阿城紫皮分蘖洋葱鳞茎茎尖愈伤组织诱导培养基以MS + 2 ,4 D 2 .0mg·L- 1+KT0 .5mg·L- 1最佳 ,出愈率为 10 0 % ;分化培养基以MS +NAA 0 .1mg·L- 1+BA 0 .4mg·L- 1最佳 ,分化率为 88.3% ,平均成苗数为 10 .2 ;生根壮苗最佳培养基为 1/2MS +PP3330 .1mg·L- 1+NAA 0 .0 1mg·L- 1+IBA 1.5mg·L- 1,生根率 10 0 % ,移栽成活率达 10 0 %。 This paper reports the studies on in vitro culture of shoot tip of tillered onion,including callus induction and differentiation,plantlet root development and transplantation.Results showed:(1)MS basal medium supplemen ted with 2,4 D 2.0mg·L -1 and KT 0.5mg·L -1 was advantageous to callus induction.The frequency of callus induction was 100%.(2)The combination of NAA 0.1mg·L -1 and BA 0.4mg·L -1 was the best for callus differentiation with the ratio of 88.3%.Most of callus was aggrandized again and induced green bud spots then formed multiplying clumpy buds,the mean number of plantlets per callus was 10.2.(3)100% of the regenerated plantlets treated with NAA 0.01mg·L -1 and IBA 1.5mg·L -1 and PP 333 0.1mg·L -1 could develop their roots,grew robustly and robust green leaves,after the regenerated plantlets were transplanted to soil.The survival ratio of transplanting reached 100%.
