大豆顶芽和子叶节组织培养再生成苗的研究 被引量:1

摘要 以大豆品种小黑豆的顶芽作外植体,研究了激素、基本培养基对愈伤组织诱导及植株再生的影响.B_5+2,4-D0.5mg/L+KT0.1mg/L 作诱导培养基,愈伤组织诱导率达92.4%;愈伤组织在 MS+6-BA1mg/L+KT0.5mg/L+ZT0.5mg/L 的培养基上,分化频率47.2%.以子叶节为外植体,通过附加外源的6-BA 刺激子叶节处潜在的分生细胞,使其不断分裂,结果子叶节膨大增粗.切下子叶节培养,结果每个子叶节最高能分化出11棵正常的苗. Using apical buds of Xiaoheidou as explants,the authors studied the effects ofhomones and basal media on the induction of calli and the regeneration of plants.The in-duction frequencey of calli on B_5+2,4-D 0.5 mg/L+KT 0.1 mg/L medium was 92.4%;When MS+6-BA 1 mg/L+KT 0.5 mg/L+ZT 0.5 mg/L was used as differentiationmedium,the differentiation frequency of buds reached as high as 47.2%.With cotyledonarynodes of Xiaoheidou as explants,by means of stimulating the cotyledonary nodes withhormone 6-BA,as many as 11 morphologically normal plants were obtained from the mul-tiple buds of each cotyledonary node.
