氨茶碱对哮喘豚鼠嗜酸细胞凋亡及bcl-2 mRNA表达的影响 被引量:11

摘要 目的观察氨茶碱对体外不同密度嗜酸细胞 ( Eos)凋亡及 bcl- 2 m RNA表达的影响 ,探讨其促进哮喘 Eos凋亡的机制。方法卵蛋白激发哮喘豚鼠动物模型 4 8h后行支气管肺泡灌洗 ,分离不同密度 Eos。氨茶碱 ( AM) ( 10 - 6 ~ 10 - 3m ol/L)干预 2 4 h,原位杂交检测 Eos的 bcl- 2 m RNA表达 ,TU NEL 法检测细胞凋亡。结果 AM干预 2 4 h,可见 Eos凋亡增加 ,以低密度 Eos( HEos)为明显 ,bcl- 2 m RNA表达降低 ,呈剂量依赖性。bcl- 2 m RNA的表达与 Eos凋亡呈负相关。结论 AM可抑制 bcl- 2 m RNA表达 ,促进 Eos凋亡。 bcl- 2参与了 AM对 Eos凋亡的调节。 ObjectiveTo investigate the effects of aminophylline (AM) on expression of bcl-2 mRNA and apoptosis in eosinophils(Eos) of bronchoalveolar lavage fluid(BALF) in asthmatic guinea pig, and to inquire into the mechanism of AM promoting Eos apoptosis. Methods 48 hours after guinea pigs were stimulated by ovalbumin(OVA), Hypodense Eos(HEos)and Normodense Eos(NEos) were purified from BALF by gradients of percoll. 24 hours after Eos cultured in PRMI1640 medium containing AM(10 -6 ~10 -3 mol/L), the mRNA of bcl-2 in Eos was measured by hybridization.Apoptosis was de- tected by TdT-mediated dUTP nick end labeling. Results Eos cultured under the condition of AM administration in vitro, apoptotic Eos increased and the expression of bcl-2 mRNA decreased in a dose-dependent manner. The expression of bcl-2 mRNA was inversely correlated with Eos apoptosis. Conclusion AM may promote Eos apoptosis and inhibit expression of bcl-2 in dose-dependent manner in vitro. bcl-2 might be involved in the mechanism that AM may promote apoptosis of Eos.
