肺癌DC融合细胞诱导的抗肿瘤免疫应答 被引量:7

摘要 目的研究肺癌 DC融合细胞 FL D- A11体内外诱导免疫应答的能力。方法用 MTT法测定 FL D- A11细胞诱导淋巴细胞增殖的能力 ;EL ISA法检测其诱导淋巴细胞分泌 IL- 2的水平 ;L DH释放法测定 FL D- A11细胞免疫后小鼠脾淋巴细胞的特异性 CTL 活性。结果 FL D- A11细胞能有效刺激淋巴细胞的增殖反应 ( SC∶ RC=1∶ 5 0时 ,SI=2 .3 8) ,诱导淋巴细胞分泌 IL- 2。 FL D- A11细胞免疫后 ,小鼠的胸腺和脾脏重量均高于对照组 ( P<0 .0 5 ) ,脾淋巴细胞对 L ewis肺癌细胞的杀伤活性显著高于对照组。结论 FL D- A11细胞具有诱导初次抗肿瘤免疫应答的能力 ,不仅能在体外诱导淋巴细胞的增殖和 IL- 2的分泌 ,而且体内免疫接种也能引起小鼠免疫器官的增生反应 ,产生针对 L ewis肺癌的特异性 CTL 杀伤作用。 ObjectiveTo explore the immunological activity of Lewis lung cancer cells and DC fused cell-FLD-A11. Methods Lymphocytes proliferation was examined with MTT assay and the supernatants was examined with the IL-2 ELISA kits.The CTL cytotoxicity was examined with LDH assay. Results Lymphocytes proliferated effectively when cultured with FLD-A11(SC∶RC=1∶50, SI=2.38)and IL-2 was secreted in the supernatant. When mice were inoculated with FLD-A11, their thymus and spleen proliferated significantly(P<0.05). The specific cytotoxicity against Lewis lung cancer cell(L3-8)was induced by FLD-A11 vaccination. It was significantly higher than the control group. Conclusions FLD-A11 cell had abi- lity to induce primary anti-tumor immune responses. It was functional in stimulating the proliferation and IL-2 secretion of lymphocyte in vitro, and CTL activity against Lewis lung cancer cell line was induced by its vaccination. The results lay an experimental foundation for FLD-A11 specific active immunotherapy.
