摘要
目的 观察氟伐他汀对高脂饮食兔动脉粥样硬化发生的早期干预作用。方法 健康纯种新西兰白兔40只 ,随机等分为实验组 (高脂饮食 +氟伐他汀10mg·kg-1·d-1)和对照组 (高脂饮食 )。分别于实验前及实验第 2、4、6、8周每组随机抽取 4只 ,兔耳中动脉抽血测血脂水平 ,气栓法处死测定主动脉内斑块面积占内膜总面积的比例。结果 实验组和对照组两组实验前的总胆固醇、低密度脂蛋白胆固醇及主动脉斑块面积占内膜总面积的比例均无差异 ,虽然随着时间的推移 ,两组的总胆固醇、低密度脂蛋白胆固醇及主动脉斑块面积占内膜总面积的比例均上升 ,但从实验第 2周以后的总胆固醇、低密度脂蛋白胆固醇及主动脉斑块面积占内膜总面积的比例治疗组均低于对照组 (P均小于 0 .0 5 )。结论 氟伐他汀能早期干预高脂饮食兔动脉粥样硬化的发生。
Objective To observe early interfering effect of fluvastatin on atherosclerosis in rabbits fed by hypercholesterol diet. Methods Forty white rabbits were randomly divided into control group (Hyperlipid diet: 1% cholesterol, 1.8% lard, 7.5% yolk powder and 83.75 % common pellet) and treated group (Hyperlipid diet+ fluvastatin 10 mg·kg -1 ·d -1 ). Four rabbits were randomly selected in each group before experiment and 2 week, 4 week , 6 week and 8 week after treatment with highlipid diet. The blood samples were drawn in order to measure serum lipids, then the rabbit was killed by gas, the aorta were isolated and handled for measuring the ratio of aortic plaque area to whole intimal area. Results There was no difference in the total cholesterol(TC)、low dendsity lipoprotein cholesterol( LDL C ) and the ratio of aortic plaque area to whole intimal area(APWIR) between the two groups before experiment. The TC、 LDL C and the APWIR were increased in the two groups as time passed in rabbits treated with hypercholesterol diet and TC、 LDL C and the APWIR in treat group are lower than in control group, there were shown to be statistical difference between the two groups after 2 weeks treatment( P <0 05). Conclusion Fluvastatin can early interfere and delay the pathogenesis of vascular atherosclerosis in rabbits fed by hypercholesterol diet.
